What the LIF format is

LIF is Leica Image File format, a proprietary container used by Leica microscopy systems and software such as Leica Application Suite, LAS X, and confocal imaging workflows. A single file can contain multiple image series, channels, z-slices, time points, and acquisition metadata rather than one ordinary bitmap.

Because LIF is a microscopy container, general image viewers may not open it correctly. Fiji/ImageJ with the Bio-Formats plugin is a common desktop option for reading LIF data and selecting a particular series or plane.

What the PNG format is

PNG is a lossless raster image format supported by web browsers, office software, image editors, and operating systems. It supports grayscale, RGB, RGBA transparency, and commonly used 8-bit or 16-bit channel data without JPEG-style compression artifacts.

PNG is suitable when the output must be a standalone image for annotation, presentation, publication drafts, or web use. It does not preserve the complete LIF container structure, microscope metadata, multiple channels, z-stacks, or time series in one normal PNG file.

How to actually convert LIF to PNG

Using the online converter

1. Open the LIF-to-PNG converter on 101convert.com; it can be used for free without installing software.
2. Upload the .lif file and start the conversion.
3. If the file contains several series or dimensions, select the required image or output option when the converter presents one. A multi-image LIF may produce several PNG files or a downloadable archive.
4. Download the resulting .png file or archive and open it in an image viewer to verify the selected frame, dimensions, and colors.

Using Fiji and Bio-Formats

1. Install Fiji, which includes the Bio-Formats importer, then open the file through Plugins → Bio-Formats → Bio-Formats Importer.
2. Select the required LIF series in the import dialog. Use options such as Split channels or Open all series only when separate channel or series images are needed.
3. For one displayed plane, choose File → Save As → PNG and specify a filename such as sample.png.
4. For every slice or time point, choose File → Save As → Image Sequence, select PNG as the format, and choose an output folder. Fiji writes one PNG per plane.

Quality, compatibility, and limitations

PNG preserves the pixel values that are exported, but it cannot preserve all information in the source acquisition. Exporting a composite RGB view discards independently editable channel data; exporting an 8-bit image also discards intensity precision from a 12-bit, 16-bit, or floating-point source.

For quantitative microscopy, keep the original LIF and export 16-bit grayscale PNGs when the receiving software supports them. Avoid screenshots because display scaling, overlays, and contrast adjustments can change the recorded values. Check the image dimensions and channel appearance after conversion, especially when the LIF contains multiple series or a nonstandard color mapping.
