What the LIF format is

LIF, usually meaning Leica Image File, is a proprietary microscopy container created by Leica Microsystems software such as Leica Application Suite and LAS X. A single LIF file can contain multiple image series with separate channels, z-stacks, time points, tiled scans, and microscope metadata.

LIF files are commonly produced by Leica fluorescence, confocal, and widefield microscope systems. They may not open correctly in ordinary photo or document applications because the image data can be multidimensional rather than a single flat picture.

What the TIFF format is

TIFF is a widely supported raster-image format used by image editors, desktop-publishing applications, scientific software, and archival workflows. It supports grayscale, RGB, and multichannel images, as well as 8-bit, 16-bit, and some 32-bit pixel data.

TIFF is suitable when you need high image quality and broad compatibility. LZW and ZIP compression are lossless; uncompressed TIFF files are larger, while JPEG-compressed TIFF can discard information and is generally unsuitable for quantitative microscopy.

How to convert LIF to TIFF

Using Fiji with Bio-Formats

1. Install Fiji, which includes the Bio-Formats microscopy importer.
2. Open Fiji and choose Plugins → Bio-Formats → Bio-Formats Importer.
3. Select the .lif file. If it contains several series, choose the required series in the series-selection dialog.
4. Keep the original bit depth. Enable options such as View stack with or Open all series only when you need a stack or multiple series in Fiji.
5. For an ordinary TIFF, choose File → Save As → Tiff and specify the output filename.
6. For a multidimensional dataset, use Plugins → Bio-Formats → Bio-Formats Exporter when available, then select TIFF or OME-TIFF and configure the required compression and dimension settings.

Fiji can also open LIF files through File → Open when the Bio-Formats reader is registered, but the Bio-Formats Importer provides more control over series, channels, and stacks.

Preserve 16-bit data for quantitative measurements, intensity comparisons, and later processing. Converting to 8-bit reduces the available intensity range and cannot be reversed. Confirm pixel size, z-step, time interval, channel labels, and calibration after conversion because TIFF export may omit Leica-specific metadata even when the visible image is preserved.
