Free online OIF to TIFF converter

Microscopy software and image-analysis systems often require TIFF instead of Olympus OIF, especially when importing images into tools without native OIF support. TIFF also provides a more portable format for editing, publication, and archival storage.

What the OIF format is

OIF, or Olympus Image Format, is a proprietary microscopy format created by Olympus FluoView and related Olympus imaging systems. It can store multidimensional image data, including channels, Z-stacks, time points, multiple series, acquisition settings, and microscope metadata.

An OIF dataset may include the main .oif file and a companion directory such as sample.oif.files. Keep the file and its companion directory together; moving only the .oif file can make the dataset unreadable or remove image planes.

What the TIFF format is

TIFF is a widely supported raster format used by image editors, publishing software, document systems, and scientific analysis applications. It supports grayscale, RGB, and high-bit-depth images and can use lossless LZW or ZIP compression.

TIFF is suitable when compatibility matters, but a conventional TIFF may not preserve every OIF feature. For multidimensional microscopy data, OME-TIFF is usually preferable because it stores standardized metadata for channels, Z-slices, time points, and image series.

How to convert OIF to TIFF

Using Fiji and Bio-Formats

1. Install Fiji, which includes the Bio-Formats importer.
2. Open the dataset with File → Open, select the .oif file, and let Bio-Formats load the companion files.
3. In the Bio-Formats Import Options dialog, choose the required series and set View stack with to Hyperstack if the file contains Z, channel, or time dimensions.
4. For a single-plane or ordinary stack, use File → Save As → Tiff. For a multidimensional dataset, use the Bio-Formats exporter or save as OME-TIFF when available.
5. Choose lossless compression such as LZW or ZIP. Do not select JPEG compression for quantitative microscopy images.

Using Bio-Formats from the command line

The Bio-Formats bfconvert utility can read OIF and write TIFF without opening a graphical interface:

bfconvert -compression LZW sample.oif sample.tif

Use an OME-TIFF output name and the appropriate Bio-Formats options when the image contains multiple channels, focal planes, time points, or series. Verify the generated file in Fiji or another Bio-Formats-compatible viewer.

Quality, compatibility, and limitations

Check the exported TIFF's width, height, bit depth, channel count, Z-slices, and time points against the source. Some exporters flatten channels or save only the currently displayed plane.

Use 16-bit TIFF for 16-bit source data and avoid converting to 8-bit unless the destination application requires it; reducing bit depth permanently removes intensity levels. Lossless compression preserves pixel values, while JPEG compression can introduce artifacts.

Standard TIFF has practical size limits around 4 GB. Large OIF datasets may require BigTIFF or OME-TIFF support in the destination application. Proprietary acquisition settings, microscope calibration data, annotations, and some Olympus metadata may not survive conversion, so retain the original OIF dataset for archival use.
