TIFF can store multiple pages or planes, so it can represent some image stacks. OME-TIFF adds standardized XML metadata for channels, dimensions, pixel sizes, and acquisition structure. TIFF compression can be lossless, such as LZW or ZIP, or uncompressed; avoid JPEG compression when preserving scientific image data.

How to actually convert ZVI to TIFF

Using Fiji and Bio-Formats

Fiji is usually the most practical desktop option because it includes the Bio-Formats importer used to read many Zeiss files.

  1. Install Fiji and update it through Help → Update if necessary.
  2. Open the file with File → Open, then select the .zvi file. Fiji should invoke Bio-Formats automatically. If it does not, use Plugins → Bio-Formats → Bio-Formats Importer.
  3. In the import dialog, select the required series and choose View stack with: Hyperstack when the file contains channels, z-slices, or time points. Disable autoscaling if the original numeric intensity range must remain unchanged.
  4. For a standard TIFF, use File → Save As → TIFF and choose a destination filename such as sample.tif. Fiji saves the currently loaded image or stack.
  5. For a metadata-aware result, use the Bio-Formats exporter when available and save as sample.ome.tiff. Verify the channel, z, and time dimensions after reopening the result.

Using Bio-Formats from the command line

Bio-Formats provides bfconvert for repeatable conversions. A basic conversion is:

bfconvert input.zvi output.tif

For an OME-TIFF output, use:

bfconvert input.zvi output.ome.tiff

Use the Bio-Formats command-line options to select a series, split dimensions, or create BigTIFF when the output exceeds the classic TIFF size limit. Check the generated file in Fiji or another Bio-Formats-compatible viewer rather than assuming that every ZVI series was exported.
