What the OIB format is

OIB means Olympus Image Binary. It is a proprietary container used by Olympus fluorescence, confocal, and other microscopy systems to store image planes, channels, z-stacks, time points, thumbnails, and acquisition metadata.

OIB files are commonly created by Olympus cellSens, Olympus FluoView, and related Olympus imaging software. You may encounter them when receiving microscopy data from an Olympus instrument or exporting an experiment from an Olympus workstation. An OIB can contain multiple image series, so it is not always equivalent to one ordinary image.

What the TIFF format is

TIFF is a widely supported raster-image format used by image editors, scientific-image software, publishing systems, and archival workflows. It supports grayscale, RGB, and high-bit-depth images and can use lossless compression such as LZW or Deflate.

TIFF is suitable when another application cannot open OIB, when individual planes must be edited, or when a broadly readable archival copy is needed. For z-stacks, time series, and multichannel data, OME-TIFF is preferable because it stores standardized microscopy metadata alongside the TIFF image data.

How to actually convert OIB to TIFF

Using Fiji and Bio-Formats

1. Install Fiji, which normally includes the Bio-Formats Importer.
2. Open the file through File → Open and select sample.oib. If Fiji asks which series to open, choose the required series.
3. In the Bio-Formats import dialog, retain the original bit depth. Use a hyperstack or virtual stack for large z-stacks and time series; avoid converting to RGB unless the destination requires it.
4. For one plane or a simple image, choose File → Save As → Tiff and save as sample.tif.
5. For a multidimensional dataset, use Plugins → Bio-Formats → Bio-Formats Exporter when available and select OME-TIFF or another TIFF output option. Check that channels, slices, and frames are included before exporting.

Using the Bio-Formats command line

The Bio-Formats command-line utility can convert a straightforward OIB dataset with:

bfconvert sample.oib sample.tif

Use the series-selection option when the OIB contains several series, for example bfconvert -series 0 sample.oib sample.tif. Export each required series separately when the target application cannot read multipage or OME-TIFF data.

Converting without installed software

Do not use this route for confidential patient, clinical, or unpublished research data unless the site’s privacy and retention terms meet your requirements.

Quality, compatibility, and limitations

TIFF conversion does not improve the microscope data; it changes the container and may change how metadata and dimensions are represented. Preserve the original OIB as the source file, then inspect the TIFF for bit depth, channel order, z-slice count, time points, scale information, and orientation.

A conventional TIFF may flatten a hyperstack, discard acquisition metadata, or save only the currently displayed plane. Exporting OME-TIFF through Bio-Formats is safer for scientific analysis because it can retain multidimensional structure and standardized metadata. Lossless TIFF compression does not reduce pixel values, while JPEG compression can introduce artifacts and should be avoided for quantitative microscopy.
